The present study aimed to investigate the effect of cryptococcal sonicated antigen and chitosan on the infection with this fungus which isolated from human skin lesions.<\/p>\n

In order to achieve\u00a0 this goal, usage of\u00a0 Cryptococcus<\/em>neoformans<\/em>isolate which was isolated from human skin lesion in order to prepare the sonicated antigen also\u00a0 60 white mice from\u00a0 both sexes, at average age 8-10 weeks were getted and divided randomly into 4 groups equally and treated as following:<\/p>\n

fifteen (15) mice were immunized with 0.5ml of WSC Ags (protein concentration14mg\/ml )S\/C two doses ,one week interval.<\/p>\n

fifteen (15) mice were fed\u00a0 diet supplemented with chitosan 1g\/kg diet for 6 weeks.<\/p>\n

fifteen (15) mice were immunized as 1st group and at the same time fed diet supplemented with chitosan as 2nd group.<\/p>\n

fifteen (15)\u00a0mice, which seted as control positive group.<\/p>\n

At day 28-30 post-immunization. skin\u00a0and\u00a0 indirect\u00a0 passive\u00a0haemagglutination\u00a0 tests\u00a0 were done ,then 1st\u00a0 ,2nd ,3rd\u00a0,and 4th groups were inoculated intraperitoneal with\u00a0 0.3 ml of fungal suspention containing 1X107<\/sup>fungal cells, all animals were sacrificed at day 30 post-infection.<\/p>\n

The study shows that the immunized animals with WSCAgs expressed high values ofdelayed type hypersensitivityand high levels of Abs titer as well as the chitosan stimulate the immune system, whileall animals of control positive\u00a0 were died at\u00a0the first week post-infection.<\/p>\n

There are\u00a0 no clear lesions in the kidney and brain and the lesions in the spleen and lung aremarked\u00a0 proliferation of\u00a0 lymphocytic cells in the periarteriolar sheath and\u00a0 proliferation of\u00a0 mononuclear cells\u00a0 around sinuses form cord like appearance\u00a0 , the liver\u00a0 showed\u00a0 mature granulomatous lesion consisting of aggregation of active macrophages in the liver parenchyma ,in\u00a0 other animals ,The liver showed multiple granulomatous lesions in portal area consisting of aggregation of epithelioid cells, macrophages ,lymphocytes around few degenerative\u00a0 yeast cells and surrounded\u00a0 by fibrous connective tissue ,the fibrosis infiltrated with\u00a0 large\u00a0 number of\u00a0mononuclear cells extended to capsular region. infected animals\u00a0\u00a0 were died during the first\u00a0 2 weeks post-infection with severe fungal cells isolated from internal organs\u00a0 and severe\u00a0 suppurative\u00a0 inflammatory\u00a0reaction in the examined organs\u00a0during early stage\u00a0 of infection\u00a0 with granulomatous lesions appear at day 14 post-infection .while no mortality\u00a0 rate was seen in animal fed diet supplement with chitosan\u00a0 with or without immunization\u00a0 moderate fungal isolation and granulomatous lesions were recorded in the examined organs of\u00a0animals sacrificed at day 30 post-infection<\/p>\n

It concluded that cryptococcal sonicated antigen\u00a0 and\u00a0 chitosan play important role in protective immunity against C<\/em>.neoformans<\/em>infection.<\/p>\n
